COREGRAPEGEN: Explo itation of the natural diversity of grape through functional genomics for improved resistance and quality

Grapevine (Vitis vinifera L.) and wine represent a relevant share of agricultural economy in Europe as well as an important part of European culture. Increasing concern on European consumers to reduce the environmental impact of agriculture and to improve food quality and safety is creating a need to improve pathogen resistance of grapevine varieties while maintaining or improving grape quality. The genetic determinants of grape quality are practically unknown in Vitis vinifera. Furthermore, genetic sources of pathogen resistance are mainly found in low quality wild Vitis species from American and Asian origin. Therefore, there is a requirement to speed up the identification of genes required for both resistance and quality traits and use this knowledge to support and accelerate the breeding of this perennial species for both table grape and wine production purposes.
Association studies based on linkage disequilibrium have been especially useful for studying the genetic basis of human diseases for which controlled genetic experiments are not feasible. They have also great potential for resolving individual genes responsible for quantitative traits.
Goal
The aim of this project is to explore the use of the genetic diversity existing in the cultivated and wild populations of grapevine in gene identification and functional analysis.

Actions
- Evaluation of the available genetic diversity in cultivated and wild genotypes and selection of a core genotypic collection for exploitation of this diversity in gene identification and functional analyses
- Molecular characterization of the nucleotide diversity in candidate genes (chosen on the basis of gene expression studies, co-location with QTLs and synteny) for quality and resistance
- Association studies between candidate genes sequence variation and traits. The project will organize data in suitable databases and adjust bioinformatic methodologies according to its requirements
- use of LD to reduce the confidence interval at a QTL. The whole project will focus on relevant traits in grapevine related with both quality and pathogen resistance.
Deliverables
As a final result the project will generate a core collection of grapevine genotypes covering a high percentage of the allelic diversity conserved in germplasm stock centres, relevant information on nucleotide diversity, an estimation of the extent of linkage desequilibrium in grapevine in specific regions investigated and the identification of the gene sequences responsible for specific quality and resistance traits in this species and the feasability of using LD-based approaches to reduce the confidence interval of QTLs.
