Description
Chinese narcissus is well-known monocot plants with beautiful color, fresh and sweet floral scent. A lack of transcriptomic and genomic information hinders our understanding of the molecular mechanisms of narcissus floral scent volatiles biosynthesis. Hence, we hypothesized the functions of the significant differentially expressed genes (DEGs) identified using Illumina RNA-Seq technology, according to public protein annotation databases in this study. Overall design: Species Jinzhanyintai of narcissus were grown in the experimental plantation of Institute of Subtropical Agriculture, Fujian Academy of Agricultural Sciences, China. Narcissus petal and corolla tissues of samples were collected at the first day of the early flowering stage (Day1) and the seventh day of the full-bloom stage (Day7) . Two petal samples (Day 1 and Day 7 stage, also short for P1 and P7) and two corolla samples (Day 1 and Day 7 stage, also short for C1 and C7) were used for transcriptional sequencing. Each sample contained two biological repeats. After collected from the plantation, sample tissues were frozen with liquid nitrogen and stored at -80°C for using. Subsequently, the eight sample tissues were entrusted to Beijing Biomarker Company (Beijing, China) for RNA extraction, cDNA libraries construction, transcriptome de novo assembly and sequencing using an Illumina HiSeq system. We named the samples as following - T01: Corolla of the early flowering stage on the first day- repeat sample 1; T02: Corolla of the early flowering stage on the first day- repeat sample 2; T05: Corolla of the full-bloom stage on the seventh day- repeat sample 1; T06: Corolla of the full-bloom stage on the seventh day- repeat sample 2; T03: Petal of the early flowering stage on the first day- repeat sample 1; T04: Petal of the early flowering stage on the first day- repeat sample 2; T07: Petal of the full-bloom stage on the seventh day- repeat sample 1; T08: Petal of the full-bloom stage on the seventh day- repeat sample 2. In this article, we mainly analyzed the genes expressed in corolla at the first day versus the seventh day (T01_02 VS T05_T06), and the genes in petal at the first day versus the seventh day (T03_04 VS T07_T08).
