Description
We have generated a chromosome-scale genome assembly of Macadamia integrifolia cultivar HAES 741, improved from the first draft genome assembly (Nock et al., DOI 10.1186/s12864-016-3272-3) by the generation of new sequencing data using a combination of high coverage short read Illumina sequences and single-molecule real-time (SMRT) long reads, and genetic linkage maps obtained from crosses using HAES 741 cultivar as a common parent. The assembled scaffolds from sequencing were anchored and ordered to the genetic linkage map to produce a chromosome-scale assembly. We also generated RNAseq transcriptome data for young leaf, shoot, and flower and used these datasets as evidences for gene prediction of the genome assembly. We anticipate that the chromosome-scale genome assembly will be an important resource for comparative genomics, association studies, and discovery of candidate genes/alleles important for macadamia improvement.
