To identify components involved in the regulation of AtALMT1 expression, we carried out a forward genetic screen on an EMS-mutagenized population in the pAtALMT1:LUC transgenic line background. hpr1-7 was identified from this screen, which showed decreased expression of the LUC reporter gene and AtALMT1. To clone the responsible gene for hpr1-7, F2 plants with decreased LUC expression from a cross between hpr1-7 and wild-type plants were pooled for DNA extraction and whole genome DNA sequencing. Through Mutmap analysis and dCAPS marker-based linkage analysis, HPR1 was finally cloned and found to be involved in the regulation of STOP1 mRNA export.
