• Description: The T-DNA insertion mutant lhr in the Hwayoung background was obtained from RiceGE (http://signal.salk.edu/cgi-bin/RiceGE/). Seeds of wild-type rice and the lhr mutant were sterilized by 0.1% HgCl2, germinated, and grown in a light chamber with the temperature of 22°C (dark, 8h) to 28°C (light, 12h). The seedlings at three-leaf stage were collected for RNA purification, and deep sequencing of the resulting RNA samples was performed by the Hanyu Corp., Shanghai, China. The normalized read count (in reads per million [RPM]) was calculated by dividing the raw read counts by the total mappable genome read counts and multiplying the resulting value by 106. The abundance of each LHR-phasiRNA was compared between WT and lhr mutant.
