Description
FUL has a dual role in regulating floral transition and fruit development. We used ChIP-seq to map and compare the FUL binding events in the two different tissues. Overall design: We used a pFUL:FUL-GFP line to conduct chromatin immuno-precipitation experiments with GFP-specific antibodies sampling two different tissue types, inflorescence meristems and pistils/siliques. The IP was followed by library preparation and deep-sequencing (ChIP-seq) to determine the FUL binding sites on a genome-wide scale. Inflorescence meristems were harvested from 4-5 weeks old 35S:AP1-GR ap1 cal pFUL:FUL-GFP lines, while stage 12-16 pistils/siliques were harvested from ful-1 pFUL:FUL-GFP lines. The tissue was fixed for 15-30’ and the immunoprecipitation was performed according to Kaufmann et al., 2010 (doi: 10.1038/ nprot.2009.244.). Experiments were done in two biological replicates with the according input sample as control.
