Identification of Heat Shock Transcription Factor (HSF) Genes Involved in Plant Thermotolerance by De novo Transcriptome Sequencing of Octoploid Cultivated Strawberry (Fragaria x ananassa Duch. cv. Toyonoka). To obtain a global overview of the strawberry transcriptome and gene activity at nucleotide resolution level, we first extracted total RNA from five different strawberry tissues including the leaves, floral buds, opened flowers, immature fruits, and roots. This preparation was for setting up the OF transcriptome dataset. Second preparation by extracting total RNA from two distinct tissues containing vegetative meristerm and inflorescence meristerm was conducted to set up the DU transcriptome dataset. Equal amounts of total RNA from each sample were pooled together, and the mRNA was isolated, enriched, sheared into smaller fragments, and further reverse-transcribed into cDNA. The different separated cDNA pools from the two preparations were subjected to Illumina HiSeqTM 2000 sequencing, and the resulting sequencing data were examined by bioinformatic analysis.
