Description
Plants uptake nitrogen (N) in two main forms: ammonium (NH4+) and nitrate (NO3-). Several ammonium transporter (AMT) and nitrate transporter (NRT) genes are deployed in uptake and transport of N-compound molecules (NH4+ amp; NO3-) from the soil into the roots and then to other plant organs. Wheat coding sequences obtained from ENSEMBL were BLAST-searched against known AMT and NRT sequences of Arabidopsis, barley, maize, rice, and wheat to retrieve homologous gene sequences in wheat. Phylogenetic relationship among these wheat genes were established using Bayesian models. Results showed distinct classification of sequences with significant homology to NRT1, NRT2, and NRT3 (NAR2) wheat sequences. Members of same gene families (NPF, NRT, NAR) also resolved into close groups. To study the expression patterns of these genes, RNA-seq was carried out on whole transcriptome of 21-days old seedlings of five spring wheat lines grown in controlled environment. Eight AMT and 52 NRT genes were differentially expressed between root and shoot tissues. Several other genes did not express neither in root nor in shoot. Seventy-five percent of differentially expressed wAMT genes and 69% of differentially expressed wNRT genes were observed in at least two out of the five wheat lines. Homeologous genes, characterized by sequence homology, revealed that their counterparts exhibited different expression patterns. Gene ontology classification of the genes showed that most were involved in several cellular and intracellular processes such as binding, catalytic activity, receptor and transporter activities as well as in metabolic processes.
