Description
Tomato leaf mold is an important disease caused by Cladosporium fulvum .The Cf-10 gene ,which has been shown to confer resistance to this disease in the field,can serve as a new candidate gen in breeding,but the lack of understanding about this gene restricts its application .In this study,we use the transcriptome technology to isolate tomato genes related to the Cf-10-mediated resistance response.The Solanum lycopersicum (contain Cf-10 tomato) and the Money Maker( contain Cf-0 gene) were grown in a greenhouse at the Horticultural station of Northeast Agricultural University (Harbin, China). The growing condition was 16-hour light and 8-hour darkness at 25 degree Celsius with an ambient humidity of 95 percent. C. fulvum physiological race 1.2.3.4 was acquired from tomato-growing regions in Harbin using the single sporangiophore transfer method. At the five-four leaf stage, the abaxial surface of the Cf-10 and Money Maker tomato seedlings was inoculated with a suspension of 1×107 sporangia per ml. Leaf samples were harvested at 0-21 days of post-infection (dpi) for microscopic analysis. According to the results of microscopic observation, samples were selected at 0 dpi (Cf10-0,MM-0), 16 dpi (Cf10-16,MM-16) for mRNA-Seq analysis. And select some keypoint to do the expirment qRT-PCR. Three samples were used for each time point.
