Description
Solanum chacoense is a wild diploid potato species, widely distributed from the South-Eastern regions of Peru to central Argentina. With our interest in studying pollen-pistil interaction and especially pollen tube guidance, we sequenced Solanum chacoense reproductive tissues, including ovules and pollen tubes, to provide a scaffold assembly, using both 454 pyrosequencing and Illumina Hiseq 2000 platform. Two individual assemblies were generated and a 454/Illumina hybrid assembly was created for S. chacoense following our data curation protocol.For differential gene expression analysis, we sequenced pollen tube attracting ovules (mature ovules) as well as pollen tube non-attracting ovules (immature ovules at two days before anthesis and mutant frk1 ovules that entirely lack an embryo sac). Genes that are present in attracting ovules and are low/absent in non-attracting ovules are considered to be candidates potentially involved in late stages of embryo sac maturation and in pollen tube guidance processes. Gene expression in leaf samples is also included as sporophytic expression control, as compared to ovules.
