Description
Oryza officinalis was planted at natural temperature and light in Jinghong, Yunnan Province of China. Samples of roots, stems and leaves were collected. At least three independent biological replicates of each tissue sample were harvested and immediately frozen in liquid nitrogen. Total RNA was isolated using the Trizol reagent (Invitrogen, USA) according to the manufacturer’s instructions. The RNA samples were treated with 10 units of DNase I (Fermentas, USA) for 30 min at 37 ℃ to remove the genomic DNA. The RNA concentrations were measured with Qubit fluorometer (Invitrogen, USA) and the RNA quality was verified using the Agilent 2100 Bioanalyzer with a minimum RNA integrated number value of 8.Qualified total RNA extracted from each sample were used for RNA sequencing by Hiseq 2000.
