Description
Pollination induces molecular changes that lead to senescence in petunia corollas. This RNA-seq study was conducted to identify early pollination-associated gene expression changes in Petunia × hybrida Mitchell Diploid. Corolla (petal) tissue was collected at three specific time points (12, 18, and 24 h) after flower opening from pollinated and unpollinated flowers. After RNA extraction, 18 paired-end, strand-specific libraries were created for Illumina sequencing. Three flow cells were used to generate nearly 500 M, 101 bp reads. These deep sequence reads were used to generate a de novo assembled corolla transcriptome and identify genetic changes that result from pollination. Changes in expression could be measured within just 12 h after pollination, well before fertilization, petal wilting, and ethylene production.
