Description
We investigated nucleotide diversity across the genic regions of the cultivated and wild type chickpea genome using six Cicer arietinum genotypes (desi type: ICCV 96029, ICC 12512-1 and Y9563-28; Kabuli type: Amit, CDC Frontier and CDC Xena), and two Cicer reticulatum genotypes (Cr5-10 and ILWC 118) using targeted 3-cDNA transcript profiling and 454 pyrosequencing technology. Eight individual genotype specific sequencing libraries were constructed in the following manner: Total RNA was extracted from each genotype individually at various developmental stages, including seedling emergence, 8-10 node stage, early flowering stage, early pod stage and early senescence. Equal amount of RNA of individual genotype at each developmental stages were pooled and Aci I digested 3’-anchored cDNA libraries were constructed. Each line was sequenced using the Roche 454 Titanium sequencing protocol. The project was conducted by the Pulse Research Group, Crop Development Centre, University of Saskatchewan and NRC, Saskatoon, SK, Canada.
