Description
This project utilises ChIP-seq to analyse the diel dynamics of H3K27me3 and H3K4me3 in the natural environment. Field sampling was conducted every 6 hours for 2 days in September 2016, in a natural population of perennial Arabidopsis halleri subsp. gemmifera in Hyogo prefecture, Japan. The leaf samples were cross-linked by formaldehyde in the field and transferred to the laboratory. To analyse the diel dynamics of the two histone modifications, ChIP was conducted for the samples collected at 8 time points, from 18.00 on 13 September to 12.00 on 15 September 2016, using anti-H3K27me3 and anti-H3K4me3 antibodies. ChIP DNA for each time point was sequenced by HiSeq2500 including four biological replicates.
