Description
Although SSR marker has practical effectiveness, detecting SSR regions for marker development often need to take arduous labors. We show an effective simple sequence repeat (SSR) detection method via capture sequencing with combination of microsatellite enrichment and paired-end sequencing by Illumina MiSeq. Five Myrtaceae plants of Feijoa sellowiana, Myrciaria dubia, Psidium guajava, Psidium littorale and Syzygium Samarangense were used for this approach. Additionally, optimum conditions for fragmentation of genomic DNA and integration of paired-end reads were investigated, and both species-specific and common SSR regions were identified within five Myrtaceae plants. This study would contribute to development of molecular marker for molecular assisted breeding and population genetics through an effective SSR finding method.
