Description
To understand the traits of gall tissue induced by an aphid Tetraneura nigriabdominalis on a leaf of Japanese elm tree (Ulmus davidiana var. japonica), RNA Sequencing analysis was performed with cDNAs prepared from leaf and gall tissues. In this study, the responsiveness of the gall tissue to jasmonic acid in relation to defense response was especially focused. Gall and leaf tissues were spray-treated with 1 mM jasmonic acid in 1% ethanol containing 0.1% Tween 20 or the solution without jasmonic acid for mock-treatment, harvested 1 hour and 4 hours after treatment, and were used for RNA preparation. Special care was taken to complete the sampling procedure within 3 min to avoid the production of wound-induced jasmonates.
